ABSTRACT:
Tyrosinase is the rate-limiting enzyme of melanin biosynthesis encoded by the TYR gene. Transcriptional regulation of this gene is specifically controlled by known transcription factors, including MITF, in neural crest-derived melanocytes and optic cup-derived retinal pigment epithelium cells. However, this locus is located within two other genes whose expression pattern is most divergent. Hence, the mammalian TYR locus has evolved to organize the surrounding chromosomal region adequately to allow precise gene regulation. We have been combining studies in transgenic mice, in CRISPR-Cas9 genome-edited mice and using advanced biochemistry on DNA and chromatin to investigate and eventually interpret the structure and function of the murine Tyr gene, in its endogenous context. In this webinar, I will discuss our current understanding of the structure and function of the murine Tyr locus.
